چکیده

At some point in their lives all cells need to grow and divide. In eukaryotic cells these processes require the delivery of new proteins and lipids from their site of synthesis in the endoplasmic reticulum (ER) to the cell surface. The Golgi apparatus, first visualised under the light microscope in 1898 by Camillo Golgi using a staining method he had developed to study neurons, lies on the route between the ER and other destinations within the cell. The electron microscopic (EM) studies of Felix and Dalton in the 1950s revealed that the Golgi apparatus is comprised of a series of overlapping cisternal or plate-like, membrane structures and associated 60 to 70 nm diameter vesicles. Palade and co-workers then went on to demonstrate from their studies of pancreatic cells that secreted proteins pass through the Golgi apparatus on their way to the cell surface or other destinations within the cell such as the lysosomes. This layered cisternal structure is often called a stack, and has a defined polarity, cisbeing the face accepting material from the ER, and transbeing the face where material exits for secretion.

The Golgi Apparatus in Amoebae

The appearance of enzymic activity during the development of the Golgi apparatus was studied by cytochemical staining of renucleated amoebae. In cells enucleated for 4 days, there was a great decline in size and number of Golgi bodies, or dictyosomes. Subsequent renucleation by nuclear transplantation resulted in a regeneration of Golgi bodies. Samples of amoebae were fixed and incubated for cy...

The appearance of carbohydrate-rich material in the developing Golgi apparatus of amoebae.

The silver proteinate reaction was used to stain carbohydrate-rich substances in normal Amoeba proteus and in the developing Golgi apparatus of renucleated amoebae. Normal cells contained stained material, which probably is glycoprotein, in the cell surface, cisternae at the concave pole of the Golgi apparatus, and cytoplasmic vesicles and vacuoles. Previous radioautographic studies had shown t...

Isolation of Germ Cell Golgi Apparatus from Seminiferous Tubules of Rat Testes

Intact Golgi apparatus have been isolated with good purity from rat testis by a simplified sucrose gradient technique. The procedure is inherently selective in that most of the Golgi apparatus in the isolate are from germ cells in late spermatocyte or early spermatid development. No Sertoli cell Golgi apparatus are present in the fraction. Biochemical analyses showed that the enzyme N-acetylglu...

RAF1-activated MEK1 is found on the Golgi apparatus in late prophase and is required for Golgi complex fragmentation in mitosis

Amitotically activated mitogen-activated protein kinase 1 (MEK1) fragments the pericentriolar Golgi stacks in mammalian cells. We show that activated MEK1 is found on the Golgi apparatus in late prophase. The fragmented and dispersed Golgi membranes in prometaphase and later stages of mitosis do not contain activated MEK1. MEK1-dependent Golgi complex fragmentation is through activation by RAF1...

The discovery of the Golgi apparatus.

The existence of the cell organelle which is now known as Golgi apparatus or Golgi complex, or simply as 'the Golgi", was first reported by Camillo Golgi in 1898, when he described in nerve cells an 'internal reticular apparatus' impregnated by a variant of his chromoargentic staining. It soon became clear that the newly-identified cytoplasmic structure occurred in a variety of cell types. Howe...

Isolation of the Golgi Apparatus from Plant Cells

A method for the isolation of the Golgi apparatus from stem tissues of onion is described. Preparations that consisted mainly of morphologically identifiable Golgi apparatus have been obtained. The best preparations were obtained from tissue homogenized under conditions of minimum shear, and in the presence of sucrose and certain additives which aid in preservation of the integrity of the Golgi...
